A transgenic construct based on a bacterial artificial chromosome containing the full
Mbl2
locus was engineered by inserting an IRES-Cre-polyA cassette into the last exon, along with a GFP reporter gene driven by a ubiquitous promoter (CAG) downstream of the 3’ end. Two transgenic mouse lines were generated via pronuclear microinjection and embryo transplantation, both of which expressed GFP.

The Cre expression pattern was analyzed by breeding these mice with the Cre-reporter,C57BL/6-Tg(Actb-GFP,-FusionRed)10Narl/Narl, which expresses FusionRed after Cre-mediated excision of a floxed GFP cassette. As expected, red fluorescence derived from FusionRed expression was observed only in the liver of this Cre transgenic line #11. FusionRed-expressing cell types in the double transgenic livers were further examined by fluorescence microscopy, revealing the characteristic histological configuration of hepatocytes. To quantify the percentage of cells with an excised allele in various organs, we performed semi-quantitative PCR using two sets of primers that amplify the floxed and excised alleles, respectively, in a single reaction. The data showed that 12.47%, 54.18%, 68.02%, and 68.47% of liver cells in 1-, 14-, 28-, and 56-day-old mice, respectively, expressed Cre recombinase. The increasing Cre activity over time may reflect hepatocyte maturation, adding another layer of biological insight. The PCR product corresponding to the excised allele was not detected in any other organs examined, except for the testis. These results indicate that this Cretransgenic mice are useful tools for investigating gene function specifically in mature hepatocytes, and they may offer more accurate hepatocyte-specific gene control than traditional lines such as Albumin-Cre.
