Strain Description
1.  A targeting vector was designed to insert exon 3 ofPrap1 gene with lacZ reporter linked to a flanked neomycin resistance cassette. The vector was electroporated into R1 embryonic stem cells. Correctly targeted embryonic stem cells were injected into C57BL/6 blastocysts. The Parp1 knockout strain has been backcrossed to C57BL/6J for at least 10 generations.
2.  The epithelial-specific expression of Prap1 was completely ablated in this strain.
3.  The length of the small intestine in this strain was significantly longer than that in control mice.
4.   On a normal chow diet, the phenotype of this strain is as follows,
 1) Prap1 knockout strain had adefect in lipid absorption.
 2) PraP1 knockout did not affect apoB mRNA expression, but significantly less apoB48              and apoB100 and triglycerideswere distributed to the CM/VLDL-rich fraction of Prap1            knockout plasma compared withthesame fraction from control mouse plasma.
5.  On a high-fat diet, the phenotype of this strain is as follows,
1) Gained significantly less body weight compared with control mice. The leaner phenotype       of Prap1 knockout mice was due to defect on lipid absorption.
2)  Fat mass percentage of the knockout mice was significantly reduced and both adipose          tissues (including iWAT and eWAT) and adipocytes were smaller than control.
      Significant decrease in the steady state plasma level of triglyceride and phospholipid.
3)  Feces of knockout mice contained more lipids than control mice.
4)  The lipid droplets, hepatocytes with vacuolation and hepatic triglyceride contents were s       significant reduced; manifesteddiminished high-fat diet-induced hepatosteatosis.
6.  Fertility is less efficient than control mice.
