Strain Description
1.  This mouse model is suitable for further studies of progression and therapeutic interventions of autosomal-dominant polycystic kidney disease (ADPKD).
2.  Targeting vector was designed to introduce a single loxP site and a loxP-flanked mc1-neomycine cassette into intron 30 and intron 34 of Pkd1 gene respectively. The targeting vector was electroporated into E14 embryonic stem (ES) cells derived from 129/Ola strain. The mice then backcrossed to C57BL/6J strain over ten generations.
3.  Mice homozygous for mutant alleles expressing a low level of polycystin-1 appeared normal at birth, but they exhibited distended abdomens and short stature at weaning. Kidney enlargement and massive renal cysts were observed as early as postnatal day14. Homozygotes died within postnatal 21 days.But this renal cystic disease was not found in heterozygotes.
