:
1.
The targeting construct was electroporated into JM8A3 (C57BL/6N background)
embryonic stem (ES) cells.
2.
The targeting vector was designed to substitute a Glycine to Aspartic acid (G53D)
at nucleotide 62 of exon 4. A loxP-flanked neo cassette inserted in intron 3 was
removed by bred mice carried Sox2-cre transgene .
3.
These mice were backcrossed to C57BL/6J for at least 5 generations.
