Strain Description
1.  The targeting vector contained mutant sequence in the proximal SF-1 binding site of Cyp11a1 (TAGCCTTGA was replaced with TAGaaTTGA), an insertion of the loxP-flanked neo gene in intron 1, was introduced into 129 embryonic stem cells by homologous recombination followed by blastocyst injection. The neo gene was removed by EIIa-cre-mediated recombination. One and halfloxP sites in the intron1 ofCyp11a1 were left after cre-mediated recombination. This strain has been backcrossed to C57BL/6 for more than ten generation.
2.  Homozygous mutant mice showed blunted HPA-axis stress response.
3.  This strain expressed about 7-fold less Cyp11a1 in the adrenal and testis but expressed normal amounts of Cyp11a1 in the placenta and ovary. This tissue-specific reduction did not affect basal steroid secretion but attenuated the circadian rhythm of glucocorticoid secretion.
