Strain Description
1.  The original construct was designed to achieve a functional humanization of the Apoe locus via CRISPR/Cas-mediated knock-in. The design intended to replace mouse exons 3 and 4. A human APOE4 coding sequence followed by an IRES-driven inducible Dre recombinase.An additional human APOE4 cistron regulated by Tet-On 3G promoter is inserted behind the mouseApoe gene. These two APOE4 genes mentioned above were flanked by the rox elements. A human APOE2 gene followed by 3XFlag Tag is the downstream of the rox element.
2.  The primary transcript of humanized APOE4 allele has been validated at the mRNA level by RT-PCR.
3.  Activation of Dre recombinase by tamoxifen is expected to mediate deletion between two rox elements and consequently, leading to conversion of human APOE4 expression to APOE2 expression.
