Strain Description
1.  This mouse model offers a tool for assessing potential therapeutic approaches for skeleton dysplasias related to over-activation of human FGFR3.
2.  The targeting vector was designed to replace the mouse Fgfr3 with human FGFR3 cDNA carrying the ACH mutation (FGGR3G380R) under the full control of the endogenous mouse Fgfr3 promoter, intron 1, and 5’ and 3’ untranslated regions. The ES cells derived from 129 carrying targeting vector was electroporated into C57BL/6J blastocysts. The Chimeric mice were crossed with 129/Sv. The offspring were maintained in 129/Sv background.
3.  Mice homozygous for this allele are viable but infertile.
4.  Heterozygous and homozygous mice were observed growth retardation, disproportionate shortening of the limbs, round head, mid-face hypoplasia at birth, and kyphosis progression during development, the phenotype shown in achondroplasia (ACH) patients. Premature fusion of the cranial sutures and low bone density in newborn mice was also observed. The phenotype above mentioned is dosage-dependent.
5.  Soften the food and place on the cage floor is required during maintaining this strain.
6.   The suggested breeding strategies of this strain are (A) heterozygote * heterozygote and (B) heterozygote (male) * 129 (the background strain) (female).
     (1) The offspring from strategy (A) could be used for tests or experiments.
          The offspring from strategy (B) is to be breeders.
     (2) The phenotype will be less and less significant if the heterozygous breeders are                          from  the littermates or offspring of the strategy (A) mentioned above.
7.  Due to the teeth protruding of ACH mice, their teeth need regular trimming, it is better to put the soaked food at the bottom of the cage.
