Strain Description
1.  A 4 bp substitution from “cggg” to “ttaa” in the first exon of Prkdc gene on rat chromosome 11q23 was introduced by CRISPR/Cas9 endonuclease originally in SHR embryos. This substitution generated a nonsense (taa) and theoretically null mutation at the 29th codon that resulted in the early termination in the translation process of the Prkdc protein, which is originally 4126 a.a. long.
2.  A heterozygous founder was selected for breeding with regular wild-type SD rats for multiple generations to dilute potential off-target effects and introduce genetic heterogeneity into this strain. Therefore, the foundation level ofanimals must be kept with such mating to select heterozygous carriers under the SD rat background.
3.  Homozygous rats are subsequently generated by sibling crossing of the heterozygous carriers, which appear physiologically normal; therefore, genotyping is required for every litter. While severe combined immunodeficiency (SCID) phenotype is found in homozygotes, fertility of homozygous rats has not been tested, but it can be expected as an option in subsequent colonies because wild-type or heterozygous littermates serve as excellent control cohorts matched for both age and sex to ensure stringent comparisons.
